Pathway Analysis of Genome Wide Association Studies (GWAS) Data Associated with Male Infertility

چکیده

Background: Infertility is a common condition affecting approximately 10–20% of the reproductive age population. Idiopathic infertility cases are thought to have genetic basis, but underlying causes largely unknown. However, basis male in humans only partially understood. The Purpose study understand current state research on genetics and its association with significant biological mechanisms. Results: We performed an Identify Candidate Causal SNPs Pathway (ICSN Pathway) analysis using genome-wide (GWAS) dataset, NCBI-PubMed search which included 632 GWAS 451 from PubMed server, respectively. ICSN produced three hypothetical mechanisms associated infertility: (1) rs8084 rs7192→HLA-DRA→inflammatory pathways cell adhesion; rs7550231 rs2234167→TNFRSF14→TNF Receptor Superfamily Member 14→T lymphocyte proliferation activation; rs1105879 rs2070959→UGT1A6→UDP glucuronosyltransferase family 1 member A6→Metabolism Xenobiotics, androgen, estrogen, retinol, carbohydrates. Conclusions: believe that our results may be helpful infertility. Pathway-based methods been applied datasets investigate reported some novel risk pathways. This pathway dataset suggests process related inflammation metabolism might contribute susceptibility. Our contribution operates through multiple genes inflammatory diseases interacting functional

Genome-wide Association Study to Identify Genes and Biological Pathways Associated with Type Traits in Cattle using Pathway Analysis

Extended Abstract Introduction and Objective: Type traits describing the skeletal characteristics of an animal are moderately to strongly genetically correlate with other economically important traits in cattle including fertility, longevity and carcass traits. The present study aimed to conduct a genome wide association studies (GWAS) based on gene-set enrichment analysis for identifying the ...

Genome-wide association studies (GWAS) and their importance in asthma.

Asthma is a complex disease determined by the interaction of different genes and environmental factors. The first genetic investigations in asthma were candidate gene association studies and linkage studies. In recent years research has focused on association studies that scan the entire genome without any prior conditioning hypothesis: the so-called genome-wide association studies (GWAS). The ...

A Bayesian Graphical Model for Genome-wide Association Studies (GWAS)

The present paradigm to analyze GWAS is to perform an exhausting testing of all single SNP associations with the response variable with the major drawback that the selected subset of SNPs has in general a very low predictive value. As a shift to the usual approach, we propose here a general statistical framework for GWAS based on Bayesian graphical modeling and able to: 1) Assess the joint effe...

Pathway analysis of genome-wide association studies on rheumatoid arthritis.

OBJECTIVES The aims of this study were to identify candidate single nucleotide polymorphisms (SNPs) and candidate mechanisms of RA and generate hypotheses for SNP 'gene' pathways. METHODS We used a meta-analysis dataset of rheumatoid arthritis (RA) genome-wide association studies (GWAS) which included 2,554,714 SNPs in 5,539 RA cases and 20,169 controls of European descent. ICSNPathway (Ident...

A pathway analysis method for genome-wide association studies.

For genome-wide association studies, we propose a new method for identifying significant biological pathways. In this approach, we aggregate data across single-nucleotide polymorphisms to obtain summary measures at the gene level. We then use a hierarchical Bayesian model, which takes the gene-level summary measures as data, in order to evaluate the relevance of each pathway to an outcome of in...
